I think Mysterio would be great both as a One-Shot terminal scenario villain where his mists, hypnogens, gases an cholograms play tricks on the players with fake enemies or disapearing ones but I also think he can have unique playstyle as a hero. Why a hero? Is it just an ilusion? Was he bored with being a losing villain?
Mysterio
Introductions
Quentin Beck was born in Riverside, California. Before becoming Mysterio, Beck was a special-effects wizard who worked for a major studio and had dreams to make a name for himself in the film industry. Beck began his career as a movie stuntman in Hollywood, then became interested in special effects. He became one of the most accomplished special effects designers in the industry, but due to lack of recognition, he wanted to become an actor. However, he lacked the looks and talent to be a star and the patience to be a director, and saw his career in special-effects to be a dead-end job. When a friend of his jokingly suggested that the quickest way to become famous was to take out a costumed hero, Beck realized that his expertise in illusions could make him an effective super-villain. Choosing Spider-Man, then a relatively weak and inexperienced target, Beck prepared his resources for a few months before attempting to eliminate the wall-crawler. Mysterio, his chosen identity, became one of Spider-Man's most elusive and persistent foes.
Mysterio once created a robotic team of X-men to fight Spider-man.
He also came back from hell and his mantle was worn by at least 2 other people.
Skills and gadgets
Quentin Beck does not possess superhuman abilities but he is an expert designer of special effects devices and stage illusions, a master hypnotist and magician, and an amateur chemist and roboticist. He had extensive knowledge of hand-to-hand combat techniques learned as a stuntman, allowing him to engage in combat with Spider-Man despite his foe's superior physical abilities and using his skills at misdirection as a further method of self-defense.
Mysterio's suit has personal weaponry which include many devices that aid him in his many agendas. His most striking feature, his helmet, is made of a one way plexiglass material, meaning he can see out but no one can see in. The helmet also includes an air supply to protect him from his own gases, a Sonar to navigate within his own mist cloak and a holographic projector to aid in 3D illusions that can be used to fool the weak willed. His boots contain magnetic coil springs which allow him impressive leaps as well as the ability to cling to surfaces. His most distinctive and persistent trademark however is his smokescreen cloak. Mysterio's costume contains nozzles in the boots and wrists that can release a constant stream of smoke, that shields his movements as well as enhancing his mystique. He can mix various other chemicals into this smokescreen for various effects. Such chemicals include; a gas that dulls and inhibits Spider-Man's spider-sense, a gas that causes paralysis for 30 minutes in those that breathe it in, a chemical abrasive that eats away Spider-Man's webbing, hypnogens that make those around him more susceptible to his will and hallucinogens to cause vivid hallucinations in those who experience it. A combination of the hypnogens and hallucinogens along with his holographic projectors are how Mysterio achieves most of his illusions. The costume sometimes also includes offensive weaponry, such as lasers or knockout gas nozzles in eye emblems on his shoulders, or electric coils within his cape to electrocute those who touch it. He also employs many different weapons, such as hand-held hypnotic aids or robotic drones, to further his aims in battle.

Smoke and mirrors
- All kind of illusions were used by Mysterio as well as smokescreens, halucynogens and other non offensive gases and they all are in this tree.
Practical effects
- Between illusions there come the real threat. Mysterio himself and his gadgets, traps, lasers, props and some tricks that when they come out of the mist they will surprise anyone even Spider-Man.
First tree employes stealth, gases for crowd control and illusions that can come in form of summons. Your gases works better when you are seen by the enemies and they want to come to you while your illusions work better when they come out of nowhere so when you are in stealth. Gasses either hang low at the feet but some are more like clouds of moving smoke. 3D projections for the player look yellow-orange.
Second tree is the offensive ones where Quentin himself steps into battle either with close quarter means of defense like acid spray and electric cape or he employ long range means of dispaching enemies with lightning bolts or this time damaging gases. This is also the tree where he summons his robots that in contrary to the 3D projections have no problem with dealing dmg to the enemies.

I never come alone Summon
When you arive you bring yourself and few more of you just for in case.
You project 3 realistic 3D illusionary versions of you in small arc in front of you.
This summons use taunt, move a little and pretend to fight in melee.
If done from stealth illusions will disapear after 2/3 of duration and apear in new pleace not far for another 2/3 of the duration again taunting.
You can have only one version of I never come alone at the same time. When used again previous one disapears.
I know your fears Area
Fear gas isn't anything to be scared of. What it makes you see is another thing.
Release fear gas around you from underneath your cape.
Aditional time if done outside of stealth.
Claustrophobia Area
Walls are closing in. What will you do when they will close on you?
You create a 4 reall lookin walls [not orange but semi transparent for players] around an area.
Targets inside are rooted, stuned and vulnerable. [If the walls can be unpenetrable by the enemies inside but penetrable from outside then the enemies don't have to be rooted or stuned]
If done from stealth all effects are reaplied after few seconds as the walls get a little closer.

There is always more mist.
You and your allies become stealth and deal more dmg.

Come and fight me Summon, Area
Won't you strike me when I'm in the open? You know you don't want to miss the oportunity.
Holographic version of you apears hovering over the mist. It firstly taunts enemis and after that it make them confused and they start fighting eachother with increased dmg. If done from stealth the Area of taunt is bigger and lasts longer to atract more enemies.
Cooldown
Cornered Movement, Area.
I don't like to run. But I don't have problem doing so.
You release paralitic gas around you, go stealth, do a short roll and gain big movement speed.
Paralitic gas knocks down and stuns. Makes enemies weaken if done outside of stealth.
Cooldown - short.
There is something in the mist Area, summon.
Mist, hypnogen, holograms and things can get real enough for anyone.
Realease mist in area and summon 4 ilusionary shadowy ninjas.
They do fast long dashes from one place to the other dealing dmg to those who belived in them.
Mist cause fear if it was done from stealth.
Only one version of this can be active.
Cooldown
Special concoction Area
I keep this for dire situations or for fun I don't remember which.
Release potent gas that confuses enemies.
You gain buff against anything in the area.
Big dodge and dmg buff.
If done outside of stealth gas works second time after the confusion is over this time it makes enemies scared.
Watch your step Area
This simple trick caused few heart attacks. It wasn't my intention.
Using the most realistic holoprojection with hypnogen in the wave form added to it you make the ground in selected area look like it crumbles and fall into an endless pit or lava [random] and it couldn't be more belivable.
Enemies in the area are stuned in fear.
Enemies in the area take more dmg.
Damage dealt to enemies in the area can prolong the stun.
Your fears are real Signature, Area, Summon
What happens when I combine all my skills? Beautifull things happen that's what.
Metal skeleton of a big robot attacks enemies with his long blades and laser. The mist that surounds him make him more real and causes enemies attacked by him to take more dmg from your other powers. On that skeleton your 3D hologram projectors put an amazing body [Example: T-rex covered in scars and alcers whose tail is a giant snake shooting lasers and the T-rex have big sword arms] that further frightens enemies.

Never alone
You are always acompanied by one illusionary version of yourself that walks near you and uses taunt from time to time.
You also can have two versions of I never come alone active.
From the backstage
When outside of fight you become stealth and gain bonus dmg when not targeted.
Mist-ic
Your gas powers have a chance to cause aditional effects, they stay longer and their radius is increased.
Practical Effects
Mysterio's basics are a little stronger than normal basics or have option in talents to be stronger. To give him another build option.
Stuntman's not faked punch Basic, Melee
You were a stuntmen in the past and you still remember your martial art lessons.
Brawling not fast attacks. Regen spirit.
If done outside of stealth +Dmg
First punch from stealth +Dmg bigger +Critical hit - but puts cooldown on the outside of stealth bonus.
Tranqualiser Dartgun Basic, Ranged
Small thing usefull at any ocasion. Just a little dart and it can do so much.
Not fast attacks speed.
Dmg, Chance to stun.
From stealth +Critical hit and chance to knockdown.
Acid from the eye Melee
Some time ago it was just for disolving web of some pesky hero. Now it does this.
Long for melee narrow cone.
Dmg + Dmg over time.
I bring lightning Ranged
Upgrade that was needed to counter the speed of some pesky hero. But it have other uses.
Shoot yellow lightning out of your glove. Hits one target.
Dmg, chance to stun.
Anything slayer Summon
Modified spider slayer that reacts to my every command. Version 1.5 almost ate me but this one follows my every whim.
You summon a modified spider slayer [looks different but have the same legs and walk - not neccessarily with the spider butt].
When the summon time is up spider jumps away like the normal spider slayers.
Cooldown
You can have only one version of Anything Slayer.
Choking gas Area, Ranged
I can make them scared, confused, I can make them laugh or simply choke and fall to the ground.
Your gas apears in target area choking enemies.
Dmg 2/s in area persists 2s after leaving area makes enemies weaken.
Don't touch the cape Melee, Area
When someone gets too close it's good to know how to get rid of them. High voltage in the cape can do that.
You swirl your cape around electrifying anyone around you.
Dmg + Chance to knockdown.
Drones Summon
Few flying killer machines aren't a bad thing to have. Never a bad thing.
Summon 4 small flying drones shaped like eyes. They fly for some time shooting lasers and after that they fly into enemies and explode. If they are in one of your smokes or gases they are stealthed and gain bonus dmg and your bonus "dmg when not targeted".
You can have only one version of drones up.
Spring coiled boots Travel power.
You do big jumps in your coiled boots traveling over obstacles.
1000 travel speed.
Beholder Ranged
Beauty of a laser is that it's just a light but it pack a hard punch.
You shoot a laser from your shoulder eyes.
Fast attack speed +Crit from stealth.
Mysterio Summon
Old times my old friend. We wanted to conquer another world in those days.
Summon Android that is similar to you but has flaming head.
He uses: your two basic powers, Bring the Lightning and Acid from the eye.
Big Cooldown
Knockdown gas Melee
Come too close get a bloody nose or something more sophisticate that don't involve me brusing my knucles.
You release condense fast acting gas in wide arc.
Dmg over time, knockdown, vulnerable.
Cooldown.
Toggles 3
No matter which toggle you chose you have map awareness thanks to your sonar.
Stuntmans life
This was your begining and it sticked to you the most.
+Hp +Defence +Dodge +Dmg to melee +Crit Dmg to melee
Propmaker
Most of your life you were a special effect guy and you do it spectacularly.
+Dmg to ranged powers +Dmg to tranqualiser dart, raised stun chance.
Bring the lightning arcs to nearby targets. [Not bounce but arcs from the main lightning on it's way]
Chocking gas makes enemies vulnerable as well.
Robotics
Newest of your profesions but it panned out pretty well in last years.
Anything slayer moves and attacks faster and leaves enemies bleeding with his lacered blades.
You can have 2 instances of drones out.
Mysterio [summmon] have increased dmg and his attacks can make enemies weaken or vulnerable.
Either +Xint [this is new field that you have learned] or just +summon dmg. - I don't know much about summon
Ultimate
Grand spectacle Area, Ranged
You create a 3D Coloseum/ Atrium [looks real not orange] around big area. It's all filled with mist and gas making different effects at random [stun, dmg 2/s, fear, confusion, weaken, vulnerable] you are in stealth all the time in that mist. Your melee powers gain dmg bonus and crit bonus from stealth. Lightning comes from the projectors you placed all around which combine with the gray mist and thunder sound make it look like a thunderstorm. Your illusion summons act as real things, your robotic summons are covered in illusions making them scarier and deal more dmg.
Talents
There is more powers here than normal so some of them can be done by talents. 3 gases can be variations of one power and already we have smaller number.
Change Cornered into option to teleport into one of your 3D summons.
When you die you are swaped with one of your illusionary summons with 20% hp.
- Come and fight me have a lightning spewing construct inside and it looks like it shoot lightning bolts from it hands
- Come and fight me there is no summon - you start levitating over mists that taunts and make enemies confused. After that you land dealing big dmg in the area.
- Mysterio changes into Big Mayan Statue summon.
- Your 3D summons from I never come alone are "real" and have 1hp but when one disapears another apears nearby like they are ghosts.
- You can change Drones into hired actors on a criminal path that fight for you but flee after time. They are dressed as zombies, pirats, magia, werewolf etc.
- You can change the way your Drones work. Instead of shooting lasers they fly in some distance from you in a patern [square but moving a little around it's corner - the square can rotate] sending their video feed to your monitors inside the globe. Because you see more with the cameras and wide view technology your dodge and critical hit rise as long as they are in the air. After time they explode.
